When during a woman's monthly cycle is the best time to get test, progesterone, estrogen levels checked?

Do woman on 10 to 20 mg test a week (average I assume) ever have to give blood or anything like that?
 
There are differing opinions on when to get labs done. Ladies have so many variables that guys do not have to deal with. I have asked several professionals and received different answers from each one. I recommend working with a HRT specialist.
My wife has never had high RBC, Hemoglobin, Hematocrit from her treatment
(test cyp and progesterone).
