Men in their twenties produce more testosterone with 600 milligrams of Tongkat Ali per day
People over fifty produce more testosterone if they take 400 milligrams of Tongkat Ali every day, but this dose has no hormonal effect in young men. If young men want to increase their testosterone levels, they need 600 milligrams of Tongkat Ali extract daily.

Study
The researchers, at the University of Malaya in Malaysia, gave 16 men aged 18-30 years 600 milligrams of Tongkat Ali extract every day for 14 days. The researchers used Physta, a water-based extract from Biotropics Malaysia Berhad. [biotropicsmalaysia.com]The supplementation provided 8.7 milligrams of eurycomanone daily. Eurycomanone analogues are the main active ingredients in Tongkat Ali.

16 men in a control group received a placebo.
Results
Tongkat Ali supplementation increased the concentration of free and total testosterone by 34 and 25 percent, respectively. In the supplementation group, the estradiol level rose by 51 percent.

The LH and FSH levels did not respond to supplementation. Based on animal data, the researchers had actually expected otherwise.
Mechanism
Based on their findings, the researchers speculate that Tongkat Ali may not increase testosterone levels in young men by making the testes produce more testosterone.
"While the hypothalamic-pituitary-gonadal axis is central to the production of testosterone, the impact of Eurycoma longifolia [Latin for Tongkat Ali - ed.] on this axis remains unclear due to lack of changes in LH, FSH and SHBG", they write. "Meanwhile, contrary to previous suggestions the increase in estradiol suggests that the aromatase may not be inhibited."
"A plausible alternative explanation on the increase in testosterone could be due to the direct stimulation of the adrenal gland in the hypothalamic-pituitary-adrenal axis converting testosterone precursor to testosterone and not entirely from the activation of the hypothalamic-pituitary-gonadal axis."
